Bioactive compounds: mechanisms of action and benefits for skin health
The efficacy of skincare products is largely attributed to their bioactive compounds, which exert beneficial effects through various mechanisms. These compounds, whether from plant extracts or biotechnological processes, can exhibit significant anti-inflammatory and antimicrobial properties.
Antioxidant properties and protection against environmental stressors
Many bioactive compounds act as potent antioxidants, scavenging free radicals and protecting the skin from damage caused by environmental stressors. Triplobiome™ Technology isolates endophytes that provide plants with resilience and protection against these stressors, transferring these benefits to the skin.
The first resulting ingredient of this endophyte isolation process is Shiloxome™, derived from the endophytic yeast Kwoniella mangroviensis found in cork oak. After a metabolomic analysis of the ferment used to create this ingredient Provital identified bioactive compounds such as hydroxylated C18 fatty acids, phenyllactic acid, 5-methoxyindoleacetate, and succinic acid. These bioactive compounds possess interesting bioactive properties such as antimicrobial, antioxidant, and collagen degradation inhibition. Interestingly, some of them (e.g., phenyllactic acid and some indoleacetic acid derivatives) are well-established plant growth promoters (produced by yeast – endophytes) that stimulate both rapid and long-term responses in plants by regulation of various developmental and physiological processes (Maki et al., 2022; Limtong et al., 2014). Therefore, Shiloxome™’s restructuring effect on the skin is the result of transferring a natural endophytic protective system to the skin.
On the other hand, the metabolomic analysis of Pureblome™ -a second endophyte-derived ingredient, isolated from the endophytic bacteria Bacillus velezensis found in the leaves of the hyssop – identified several bioactive compounds coming from both plant and bacterium. For instance, the peptides under 8 kDa identified, hold broad spectrum antioxidant properties. Surfactin, a lipopeptide also identified, shows significant antioxidant activity too, reducing lipid peroxidation and protecting against oxidative stress (Islam et al., 2022; MM Khan et al., 2020; Suchi et al., 2023).
Hydration and barrier function enhancement
Maintaining optimal skin hydration and a robust barrier is crucial. Certain bioactive compounds, including those found in plant extracts and produced by endophytes, can help retain water and support the skin’s lipid barrier.
A clear example is Shiloxome™’s effect in fortifying the skin’s microbiome barrier and reinforcing the integrity of the lipid barrier, ensuring improved moisture retention and proper epidermal function. Lipidomic analysis revealed that Shiloxome™ regulates hundreds of lipids, significantly increasing its chain lengths. This is particularly true for ceramides, which are strongly associated with a robust skin barrier function. An in vivoenrichment analysis showed that Shiloxome™ increases the total chain length of the lipidome.
In the case of Pureblome™, a specific bioactive lipopeptide (Fengycin) was identified in the ferment used for the creation of the ingredient. This compound is directly associated with anti-fungi and skin barrier strengthening. (Vanittanakom et al., 1986).
Anti-inflammatory and skin-soothing effects
Inflammation is a key factor in various skin issues, and several bioactive compounds can help mitigate these issues through their anti-inflammatory properties.
As explained above, the metabolomic analysis of Shiloxome™ identified phenyllactic acid, which has antimicrobial and antioxidant properties. Shiloxome™, in vitro, showed a dose-dependent reduction of 5 inflammation mediators (CCL5, COX2, CSF2, GRO1 and TNFA).
In this regard, Pureblome™ was also proven to offer anti-inflammatory effects thanks, in part, to the small peptides (under 8 kDa) and the Dihydroxy-eudesmanolide, both identified through metabolomics, which hold anti-inflammatory properties (Padilla-González et al., 2016); as well as the lipopeptide surfactin, that reduces inflammation in sensitive or damaged skin (Zouari et al., 2016). Besides looking at the bioactive compounds found in its ferment, Pureblome™ achieves “the right balance” with its proven anti-inflammaging action. Invitro, for instance,it reduced the secretion of pro-inflammatory cytokines IL-6 and IL-8 in sebocytes treated with linoleic acid.
Advances in bioactive compounds for skin aging
The quest for youthful skin has driven significant research into bioactive compounds that can address the signs of aging.
Stimulating collagen and elastin production
Bioactive factors can stimulate fibroblasts to produce collagen and elastin, the skin’s structural proteins.
In the case of Shiloxome™, succinic acid was identified in the previously-mentioned metabolomic analysis. This bioactive compound has properties related to collagen degradation inhibition (Maki et al., 2022). In turn, the treatment on human dermal fibroblasts showed an enrichment of pathways related to the extracellular matrix remodeling and an improvement in dermal collagen and elastin content. A global transcriptome enrichment analysis indicated that Shiloxome™ triggers pathways related to extracellular matrix structure and organization.
Pureblome™ has demonstrated the ability to stimulate collagen and elastin production by revitalizing fibroblasts and decreasing glycation in vitro. Interestingly, the lipopeptide surfactin, identified in its ferment is also known to stimulate collagen and inhibit matrix metallopeptidases (Bueno-Mancebo et al., 2024).
Reducing wrinkles and fine lines
By promoting collagen synthesis and inhibiting enzymes that degrade it, certain bioactive compounds can help reduce the appearance of wrinkles and fine lines.
Shiloxome™ also contributes to a smoother skin microstructure, with an isotropic orientation of the skin surface being statistically significant at day 56 of in vivo testing.
In vivo studies have shown that a 2% Pureblome™ formulation significantly reduced wrinkles after only 14 days, as observed through profilometry analysis. In this case, the lipopeptide mentioned above is also linked to wrinkle reduction (Bueno-Mancebo et al., 2024).
